HPLC techniques for proteomics analysisça short overview of latest developments

چکیده
Due to the complex nature of the proteome, instrumentation and methods development for sample cleanup, fractionation, preconcentration, chromatographic separation and detection becomes urgent for the identification of peptides and proteins. Newly developed techniques and equipment for separation and detection, such as nanoHPLC and multidimensional HPLC for protein and peptide separation, enabled proteomics to experience dynamic growth during the past few years. In any proteomic analysis the most important and sometimes most difficult task is the separation of the complex mixture of proteins or peptides. This review describes some aspects and limitations of HPLC, both multidimensional and one-dimensional, in proteomics research without attempting to discuss all available HPLC methods, which would need far more space than available here.

متن کاملNew Developments in Arterial Spin Labeling
Arterial Spin Labeling is a magnetic resonance method for the measurement of cerebral blood flow. The perfusion signal is usually coming from the subtraction of two successively acquired images, one with, and one without proximal labeling of arterial water spins. Over the last decade, the method has moved from the experimental laboratory to the clinical environment. Furthermore, numerous improv...
متن کاملProtein digestion: an overview of the available techniques and recent developments.
Several proteomics approaches are available that are defined by the level (protein or peptide) at which analysis takes place. The most widely applied method still is bottom-up proteomics where the protein is digested into peptides that can be efficiently analyzed with a wide range of LC-MS or MALDI-TOF-MS instruments. Sample preparation for bottom-up proteomics experiments requires several trea...
متن کاملOpenMS – A Framework for Quantitative HPLC/MS-Based Proteomics
One of the main goals of proteomics research is the discovery of novel diagnostic markers and therapeutic targets. Currently, mass spectrometry is the main platform for analyzing complex protein samples. Lately, HPLC/MS-based approaches have gained considerable interest due to their larger potential for full automation when compared to gel-based techniques. Particularly, multi-dimensional HPLC-...

Widespread studies on tumor diagnosis has led to the development of techniques with greater sensitivity and improved specificity for detection of occult and small subclinical tumors. Tumor imaging by radiolabeling of tumor-specific antibodies (Immunoscintigraphy) or tissue specific biochemical agents (Receptor imaging) and especially physiologic imaging with positron emitters (PET), have ...
